Disclaimer: The fact that this page reports negative results with the given antibody does not mean that the antibody does not work – it just means that our laboratory did not succeed, using our typical experimental protocol, to find the searched protein by immunoblotting in the tissue examined. Modificiation of the protocol, or the selection of other tissue, might still provide excellent results.

The antibody is raised against the extracellular part of matriptase-2; the datasheet does not mention whether this includes the active site (mutated in mask mice).  The antibody detects, in liver plasma membrane-enriched fraction, an extremely strong band at about 66 kDa (which could theoretically be a cleaved matriptase-2), and several weaker bands, one of them at approximately 125 kDa (which could be the full length matriptase-2). However, the strong band at 66 kDa is only very sligthly shifted by PNGase F treatment – which is in contrast with the reported extensive glycosylation of matriptase-2 (confirmed experimentally by the Abcam Ab56182 antibody). There is no difference in the 66 kDa or 125 kDa between wild type and mask mice.

In summary, we regard detection of matriptase-2 by this antibody as problematic, although definite proof would require testing in Tmprss6 -/- mice, rather than in mask mice.
